ABSTRACT

Interest to develop new anticancer drugs and to design combination treatments with little or no secondary effects provides new scope for traditional phytochemicals in chemoprevention and therapy. Propolis is a known source of polyphenols, and flavonoids found in them have been widely studied as biochemical markers for botanical origin and in explaining their antioxidant capacity as a key factor in chemoprevention. Antimicrobial, anti-inflammatory and anticancer biological activities of propolis are known. Studies of cancer cells to measure the anticancer effect of propolis are designed with one carefully chosen component, and with extracts applied to cells in culture media. The antitumor effect of propolis and caffeic acid phenethyl ester (CAPE), bioactive compound of propolis extract, is seen to be associated with its ability to initiate apoptosis of cancer cells. Chrysin is a flavonoid of interest to identify signaling molecules related to cancer. As cancer cells develop multidrug resistance (MDR) during chemotherapy, this opens a new avenue of research on cellular mechanisms of propolis components in combined treatments designed to overcome MDR. ABSTRACT

The Brazilian Sac Brood is a disease that affects apiaries of Africanized bee hives in Brazil, thereby making them susceptible to high losses. This study investigated the pathogenicity of Africanized bee hives by the entomopathogenic fungi in a Brazilian Sac Brood endemic region. The degree of fungal contamination, presence of mycotoxins in beehive elements, and vulnerability of healthy beehives in environments subjected and not subjected to the disease were investigated. From the contaminating fungal load, species that are mycotoxin producers and pathogenic causing mortality in the bees have been isolated. The analysis of bee pollen and bee bread samples did not show the presence of the toxic pollen of Stryphnodendron (Fabaceae), which has been indicated as the causative agent of mortality in pre-pupal stage larvae. A cria ensacada brasileira é uma doença que afeta apiários de colmeias de abelhas africanizadas no Brasil, tornando-os suscetíveis a perdas elevadas. Este estudo investigou a patogenicidade de fungos entomopatogênicos em colmeias de abelhas africanizadas de uma região endêmica de cria ensacada brasileira. O grau de contaminação fúngica, a presença de micotoxinas em elementos colmeia e a vulnerabilidade das colmeias saudáveis em ambientes sujeitos e não sujeitos à doença foram investigados. A partir da carga fúngica contaminante, espécies produtoras de micotoxinas e patogênicas, que provocam a mortalidade de abelhas, foram isoladas. A análise do pólen e do pão de abelha não demonstrou a presença do pólen tóxico de Stryphnodendron (Fabaceae), que tem sido apontado como agente causador da mortalidade de larvas em fase de pré-pupa. No entanto, o pão de abelha foi o substrato mais correlacionado com a contaminação fúngica...

ABSTRACT

Dengue hemorrhagic fever (DHF) is a severe febrile disease, characterized by abnormalities in hemostasis and increased vascular permeability, which in some cases results in hypovolemic shock syndrome and in dengue shock syndrome. The clinical features of DHF include plasma leakage, bleeding tendency and liver involvement. We studied the histopathological features of a fatal case of dengue-3 virus infection. The patient, a 63-year old male, presented with an acute onset of severe headache, myalgia and maculopapular rash. Tissue fragments (liver, spleen, lung, heart, kidney and lymph nodes) were collected for light microscopy studies and stained by standard methods. Histopathology revealed severe tissue damage, caused by intense hemorrhage, interstitial edema and inflammation. Some tissue sections were also processed with the immunoperoxidase reaction, which revealed the dengue viral antigen. Dengue-3 virus was isolated and identified with electron microscopy in a C6/36 cell culture inoculated with the patient's serum. Viral particles were detected in the infected cell culture.

ABSTRACT

Hepatic viscerotomy of paraffin-preserved old specimens, collected in the period from 1934 to 1967, were analyzed by immunohistochemical assays to detect hepatitis B, hepatitis D, dengue and yellow fever virus antigens. The material belongs to the Yellow Fever Collection, Department of Pathology, Instituto Oswaldo Cruz, Rio de Janeiro, Brazil and the cases were diagnosed at that time according to clinical aspects and histopathological findings reporting viral hepatitis, yellow fever, focal necrosis and hepatic atrophy. From the 79 specimens, 69 were collected at the Labrea Region and the other 10 in different other localities in the Amazon Region. The five micra thick histological slices were analyzed for the presence of hepatitis B surface antigen (HBsAg) and hepatitis B core antigen (HBcAg) by immunoperoxidase technique. An immunofluorescence assay was applied to the detection of hepatitis D, yellow fever and dengue virus antigens. Nine (11.4 percent) histological samples were HBsAg reactive and 5 (6.3 percent) were HBcAg reactive. The oldest reactive sample was from 1934. Viral antigens related to the other pathologies were not detected in this study. Our results confirm that the methodology described may be used to elucidate the aetiology of hepatitis diseases even after a long time of conservation of the specimens

ABSTRACT

A presença de antígeno viral em cortes de tecidos humanos fixados em formol e emblocados em parafina foi demonstrada pela digestäo com tripsina foi demonstrada pela ingestäo com tripsina seguida de imunofluorescência direta ou indireta. Os espécimens podem ser utilizados para diagnoses retrospectivas. A técnica da imunofluorescência deve ser adaptada à infecçäo viral suspeita segundo diagnosie histopatológica prévia. Os parâmetros para a digestäo do tecido pela tripsina, relacionados à concentraçäo, duraçäo de atuaçäo e temperatura, expöem diferentes antígenos virais e devem ser previamente testados para cada sistema a ser estabelecido. Uma digestäo mais intensa deve ser aplicada para a detecçäo do vírus do sarampo em tecido pulmonar do que para adenovírus ou vírus respiratório sincicial no mesmo tecido. Por outro lado, o vírus da febre amarela em tecido de fígado necessita de uma digestäo mais fraca

ABSTRACT

Isolamento e caracterizaçäo antigênica do vírus da imunodeficiência humana (VIH/HIV) no Brasil - Um retrovírus foi isolado de um paciente brasileiro com "Síndrome de Imunodeficiência Adquirida" (SIDA/AIDS) e caracterizado em termos de sua reatividade com soros de indivíduos infectados com vírus da imunodeficiência humana dos tipos 1 e 2 (HIV-1 e HIV-2). A análise antigênica por "Western blot" revelou que o isolado brasileiro é bastante similar a uma cepa de HIV-1 bem caracterizada. A identificaçäo do retrovírus como HIV-1 e näo HIV-2 é reforçada pelo fato dos anticorpos do paciente do qual foi isolado o vírus näo terem reagido com a glicoproteínas de envelope de 140 kDa, específica para HIV-2

ABSTRACT

Suspensoes virais de Rotavirus, Adenovirus Influenza, Parainfluenza, Virus Respiratorio Sincicial, Varicela, Caxumba, Rubeola Herpes Hepatite B, Astrovirus, Poliomielite e Vaccinia foram examinadas em microscopia eletronica diretamente ou atraves da imuno-microscopia eletronica, a fim de determinar a melhor substancia contrastante negativa especifica para cada um dos seus tipos morfologicos virais. Usando acido fosfotungstico (PTA) como contrastante padrao, os resultados foram aprimorados com o emprego do silico - tungstato de sodio (SST) para os Myxovirus, Adenovirus e virus da Poliomielite, do acetato de uranila (UA) para a Hepatite B e do citrato de uranila (UC) para os envoltorios do tipo morfologico da varicela e de detalhes da estrutura de outros tipos virais. Outrossim, suspensoes virais nao purificadas, ao serem contrastadas com molibdato de amonia (AM) e SST evidenciaram melhor as particulas virais integras. A fim de obter um melhor espraiamento das suspensoes virais, especialmente as obtidas de gradientes de sacarose e cloreto de cesio sobre as grades revestidas de Formvar e carbono, a poli-L-lisina mostrou-se superior as demais substancias testadas; alem de separar os agrupamentos virais das impurezas na imuno - microscopia eletronica, tambem permanencia aderido um numero maior de particulas virais isoladas

ABSTRACT

A evidenciacao da presenca de acido ribonucleico (ARN) viral por eletroforese em gel de policrilamida (EGPA) foi comprovada como um metodo altamente sensivel e rapido para o diagnostico de infeccoes por rotavirus. Uma comparacao desta prova com a imunomicroscopia eletronica (IEM) e com o ensaio imunoenzimatico (EIE) no exame de 245 fezes de criancas com gastroenterite revelou completa concordancia entre os tres ensaios em 238 (97.14%) amostras. Entre 75 amostras positivas pelo menos em um dos tres ensaios, resultados negativos foram observados em 5 (6.48%) por EGPA, em 6 (6.76%) por EIE e em nenhum por IEM. Coloracao pela prata aumentou consideravelmente a sensibilidade do ensaio por EGPA.Concluimos que embora a IEM ainda seja a prova mais sensivel e rapida para o diagnostico de infeccoes por rotavirus, o ensaio por EGPA tem muitas vantagens em seu favor, sendo as principais as de nao necessitar equipamentos caros, de empregar exclusivamente reagentes quimicamente definidos, de identificar grupos e variantes virais e de detectar amostras que nao possuindo antigenos comuns a maioria dos rotavirus, nao sao revelaveis por ensaios sorologicos
